Lietzow is located on Rügen at the narrowest point of the land bridge between the Großer Jasmunder Bodden and the Kleiner Jasmunder Bodden. The municipality in Mecklenburg-Western Pomerania is located approximately eleven kilometers northeast of Bergen auf Rügen.

The B96 road and the Stralsund–Sassnitz and Lietzow–Binz railway lines run directly through Lietzow. Regional trains generally continue alternately toward Sassnitz or Binz. This makes Lietzow a transfer point in the rail network.
